Design: The libraries were sequenced on the Illumina sequencing platform by Genedenovo Biotechnology Co., Ltd (Guangzhou, China). Sequencing was performed using the Illumina Novaseq 6000 platform according to the manufacturers instructions. The sequencing reads were aligned using Bowtie 2.

Streptococcus suis is a Gram-positive pathogen causing septicemia and meningitis in pigs and humans. We previously found that sRNA rss04 contributes to meningitis induction by regulating capsule synthesis and inducing biofilm formation in a mouse infection model. However, its targets are unknown. In this study, we employed MS2-affinity purification coupled with RNA sequencing (MAPS) to identify the targets of rss04.
Sample: The WT strain containing pSET2-MS2-negative (used as a negative control) were grown in 1200 mL THB medium to stationary phase. Subsequently, strains were applied to MS2-affinity purification coupled with RNA sequencing (MAPS).
